Lake Pontchartrain Basin Restoration Act of 2000 - Amends the Federal Water Pollution Control Act to add Lake Pontchartrain Basin, Louisiana and Mississippi, to the list of estuaries given priority consideration for inclusion in the national estuary program.

Requires the Administrator of the Environmental Protection Agency (EPA) to establish the Lake Pontchartrain Basin Restoration Program within EPA. Describes duties of the Administrator under such Program, including to: (1) provide assistance and support to the management conference convened for the Basin under the national estuary program; (2) support environmental monitoring of the Basin and research; (3) develop a comprehensive research plan to address Program technical needs; (4) coordinate related grant, research, and planning programs; and (5) collect and make publicly available information relating to Basin environmental quality.

Authorizes the Administrator to make grants for: (1) restoration projects and studies recommended by a management conference convened for the Basin; (2) public education projects recommended by such conference; and (3) the inflow and infiltration project sponsored by the New Orleans Sewerage and Water Board and Jefferson Parish, Louisiana. Authorizes appropriations.

Expresses the sense of Congress that all recipients of grants pursuant to this Act shall abide by the Buy American Act. Requires the Administrator to give notice of Buy American Act requirements to grant applicants.
